The interaction between theseproteins was also intricated in the GO terms and KEGG pathways,such as the positive regulation of cellular metabolic process(GO:0031325), regulation of cell proliferation (GO:0042127),protein binding (GO:0005515), enzyme binding (GO:0019899), nuclearpart (GO:0044428), microRNAs in cancer (05206), FoxO signalingpathway (04068), indicating that eleutheroside B1 can affect cellfunction by regulating both ncRNA and protein. Combining theRNA-seq and iTRAQ results, it was demonstrated that eleutherosideB1 regulated some pathways, which were associated with cellproliferation and gene expression at different levels to inhibitthe replication of influenza virus.


Further proteomics analysis of the function ofeleuthero-side B1 was performed through iTRAQ assays. Only oneprotein (LAGE3) was downregulated in the PR8+eleu vs. PR8 groups,whereas 70 were upregulated and 5 proteins were downregulated inthe PR8+eleu vs. cell groups. LAGE3 belongs to the NY-ESO genefamily, a component of the EKC/KEOPS complex. This complex isrequired for the formation of a threonylcarbamoyl group onadenosine at position 37 (t6A37) in tRNAs that read codonsbeginning with adenine (55,56). Some researchers have reported thatthe EKC/KEOPS complex is linked with the protein synthesismachinery and cell growth (57).According to GO analysis, LAGE3 was involved in tRNA processing,tRNA metabolic processes and lncRNA processing. Thesedifferentially expressed proteins could be potential targets ofeleutheroside B1 in regulating the ncRNAs of host cells. TransferRNAs (tRNAs) are substrates for protein synthesis and a centralpart of the translation machinery in all living organisms, whichimpact translational speed and fidelity by their abundance. A'channeled tRNA cycle' includes the following: tRNAs are shuttleddirectly from the ribosome to their cognate tRNA synthetase andback to the ribosome for another cycle of translation withoutreentering the cytosolic pool (58,59). The polysome-associated tRNApopulation changes dramatically, following influenza virusinfection; however, the total cellular tRNA population remainsunaltered (60). This proves thatthe changes in polysome-associated tRNA levels reflect the codonusage of viral genes, suggesting the existence of local tRNA poolsoptimized for viral translation and enhanced translationalefficiency of viral genes. Over the past decade, ncRNAs (microRNAs,lncRNAs and vtRNAs) have been identified as an important class ofregulators involved in virus-host interaction, particularly inanti-viral immune response (20).Focus on ncRNAs has recently been increased. Advances in thehigh-throughput sequencing techniques are generating increasingnumbers of newly discovered ncRNAs that are involved in infectionsand immunological processes (61). In some reports, ncRNAs have beenshown to regulate histone modifications in influenza virus-infectedcells, interact with transcription factors during virus infectionand induce specific protein-coding gene expression, affect virusreplication in an interferon-independent manner and control proteinsynthesis during influenza virus infection (62-66). The LAGE3 gene involved in lncRNAprocessing warrants further investigation to determine the role ofeleuthero-side B1 as a lncRNA regulator for its anti-influenzavirus.


Overall, the present study demonstrates that ncRNAsare involved in the interaction between host and influenza virus.Although research on non-coding RNA is currently unpopular inChinese medicine, it is worth noting that ncRNAs, such as NEAT1,the protein LAGE3, and their pathways identified in the presentstudy indicate the need to investigate further the details of theability of eleutheroside B1 to inhibit influenza virus. One of thelimitations of the present study was that it was only performedusing A549 cells. It would be ideal to include animal or humansamples to profile the RNA and protein expression, particularly ininfluenza virus-infected patients treated with this drug to comparewhether the observed changes are comparable in a clinical setting.Unfortunately, the existing method for extracting eleutheroside B1does not yield sufficient compounds to allow animal experiments andclinical tests. However, in the future the authors aim to studyclinical samples treated with herba sarcandrae. In addition,another limitation is that ncRNA expression was not examined atdifferent time points. Previous research (46) has often selected 24 h afterinfection for RNA-seq; therefore, this time point was used fordetection in this study. In the future, the authors aim to performfurther studies using different time points. Another limitation ofthe present study is that only the iTRAQ assay was used toinvestigate protein expression, which yields incomplete data due tothe use of data-dependent acquisition that only captures onedifferentially expressed protein. Thus, it will be interesting toexamine protein expression profiles induced by eleutheroside B1 byother proteomics methods, being cautious that ncRNAs and proteinsmay function in a coordinated fashion, and therefore, the overallanti-influenza virus response induced by eleutheroside B1 may bedue to the effect of a cumulative response to overall changes inncRNAs or proteins rather than individual ncRNAs or proteins.Furthermore, certain studies open novel avenues to further explorethe molecular pathways based on ncRNA or protein functions.Therefore, whether these networks were induced by eleutheroside B1in the future should be investigated.

Objective. To assess the current clinical evidence of manufacturedChinese herbal formulae (MCHF) for knee osteoarthritis (KOA). Methods. Sevendatabases were searched from inception to May 2015. Eligible randomizedcontrolled trials investigating the effectiveness of MCHF for KOA wereincluded. Data extraction, methodological assessment, and meta-analyses wereconducted according to the Cochrane standards. Results. A total of 17 kindsof MCHF were identified from the twenty-six included trials. Meta-analysesshowed that MCHF significantly relieved the global pain of knee joints,either used alone or combined with routine treatments. Additionally, MCHFplus routine treatments significantly decreased the scores of WOMAC andLequesne index. However, there were no statistical differences between MCHFgroup and routine treatment group in walk-related pain and WOMAC scores. Nosignificant differences were found in Lysholm scores. There were twenty-onetrials that mentioned adverse events. A pooled analysis showed that adverseevents occurred more frequently in control group compared with MCHF group.Conclusions. Our results indicated that MCHF showed some potential benefitsfor KOA. However, we still cannot draw firm conclusions due to the poormethodological quality of included trials.
